Footage emerged on social media this week of the Tigers superstar appearing to grab the breast of a topless woman.
The woman is sitting on the lap of another man, before Martin appears to reach around and touch the woman, to laughter of others at the bar.
When the video was filmed remains unclear, but it is believed to be several years old.
Richmond coach Damien Hardwick was asked about the vision on Wednesday but said he didn't know anything about it.
An AFL spokesperson confirmed to AAP the league was aware of the video and would be seeking "further information".
AFL chief executive Gillon McLachlan said he only found out about the video late on Wednesday night.
"At first pass, the video, it's a poor look," he told the Seven Network at Melbourne Airport on Thursday.
"I don't have any further information this morning about Dustin's relationship to the woman, her views, circumstances, the facts.
"It's with our integrity guys and they'll follow up at the right time, and I'm sure they'll try and speak to everyone involved."
But Richmond senior advisor Neil Balme downplayed the incident and said there was never any chance Martin would miss Thursday night's elimination final against Brisbane.
"I don't think we took it that seriously but we'll see what happens," he told Channel 10.
"It seemed a long time ago what you're talking about.
"We're pretty good at focusing on what needs to be focused on and the games the only thing that's important to us."
The Tigers battle the Lions at the Gabba to open this year's finals series.
Martin will play his first game since round 16 after struggling with a hamstring issue.
